Physiol, 2 I I (3) : 746-754. rg66.-Studies of membrane impedance and potential variation were made on squid giant axons under sodium-free external conditions. Intracellular perfusion by the double-cannulation method was employed. It was found that with favorable internal conditions, excitability could be maintained with several organic and inorganic sodium substitutes, and that the excitation process in sodiumfree media is not dissimilar from that in sodium-containing media. Tetrodotoxin was found to suppress excitability in all cases. The implications of these findings for the concept of sodium-specificity are discussed.
